It's worth noting that MaxOutputBufferSize setting of 0 isn't what it looks like - it actually defaults to 1024 which produces noticable audio lag. While you're tweaking MaxOutputBufferSize values, if there's crackling on the loading screen you can just kill the game with Alt-F4 and try again with a higher setting. Good luck! Update 6/2013:

There's an inherent delay in the Rocksmith RealTone cable to do some ADC (analog-digital conversion), frequency analysis, etc of your guitar's output and for the Rocksmith software to interpret it, run it through its software mixer (the pedals, amps, etc you have set up), and then send the audio out. Some of this delay is purely in detection for the purposes of checking your play …

The reson for this is that sound input that is processed needs ASIO technology for minimal lag but the game does not provide support for this. You either need a professional sound card for ASIO or you need two soundcards and use Asio4All for guitar input with minimal lag. That is if Rocksmith supported audio input through ASIO.

Windows PC are not very good with audio. there's a reason why mac is dominant in the audio world and rocksmith runs smooth no issues with it. The problem is the buffer. you would do much better to change the on-board audio driver. for pc i would recommend installing ASIO4ALL and then adjust the buffer size.

USB soundcards can cause latency, try an internal soundcard. Reduce the audio latency slider in the game's options. In Control Panel-->Sound, Set the default format for your default output device AND the rocksmith cable to 16 bit, 48000 hz (DVD Quality), as per this link: http://forums.ubi.com/showthread.php/802854-Rocksmith-2014-PC-Configuration-and-FAQ.
